VIP | Vasoactive Intestinal Peptide — C-Terminally Amidated Neuropeptide
5 mg Lyophilized Powder CAS 40077-57-4 28 AA
Receptor Target
VPAC1 / VPAC2
Research Pathway
Gαs — cAMP / PKA
Origin
Neuroendocrine / Endogenous

Vasoactive Intestinal Peptide (VIP) is a 28-residue C-terminally amidated neuropeptide first isolated from porcine duodenum by Said and Mutt in 1970. It belongs to the secretin/glucagon superfamily and shares substantial sequence homology with PACAP, with which it shares receptor targets. VIP signals through the class B G-protein-coupled receptors VPAC1 and VPAC2, coupling predominantly to Gαs and elevating intracellular cAMP. It is distributed widely across enteric, central and peripheral neurons, and is among the most extensively studied neuropeptides in smooth-muscle, circadian and immunological research models.

Research Targets & Proposed Mechanisms
VPAC1
Class B GPCR with high VIP affinity; reported in pulmonary tissue and T-lymphocyte populations. Studied in epithelial and immune cell signalling assays.
VPAC2
Second class B GPCR target; reported in smooth muscle, mast cells and basal lung mucosa. Investigated in smooth-muscle relaxation and secretory model systems.
Gαs / cAMP / PKA
Canonical downstream cascade: receptor activation stimulates adenylyl cyclase, raising cAMP and activating protein kinase A. Standard readout in receptor pharmacology assays.
Shared receptor pharmacology: VIP and PACAP bind VPAC1 and VPAC2 with comparable affinity, while PACAP additionally binds the PAC1 receptor, at which VIP is substantially weaker. That selectivity difference is routinely exploited in receptor-mapping experiments to distinguish VPAC-mediated from PAC1-mediated responses, which is a large part of why VIP is used as a reference ligand in this field.
Research Context
  • →First isolated from porcine small intestine by Sami Said and Viktor Mutt in 1970; originally characterised by its vasodilatory activity, from which the name derives
  • →Used as a reference ligand in VPAC1/VPAC2 receptor pharmacology, including competitive binding, cAMP accumulation and receptor-subtype selectivity assays
  • →Studied extensively in circadian biology; VIP-expressing neurons of the suprachiasmatic nucleus are a long-standing model for intercellular synchronisation of circadian oscillators
  • →Investigated in immunological research for proposed effects on T-cell differentiation, regulatory T-cell populations and cytokine profiles in in vitro model systems
  • →Examined in enteric neuroscience as a principal non-adrenergic, non-cholinergic mediator in gastrointestinal smooth-muscle preparations
  • →The synthetic form, aviptadil, has been developed and investigated clinically in several jurisdictions, providing an extensive translational data set alongside the preclinical literature
Storage & Handling

Store sealed lyophilized material under controlled laboratory conditions in accordance with validated institutional SOPs and applicable technical documentation. Protect from light and moisture.

  • →Keep in original sealed vial until use in research protocols
  • →The sequence contains a single methionine residue (Met¹⁷), which is susceptible to oxidation; minimise exposure to air and oxidising conditions during handling
  • →Asparagine and glutamine residues render the peptide susceptible to deamidation under extremes of pH; buffer selection should be documented in the protocol
  • →The lyophilized material is hygroscopic; allow vials to reach ambient temperature before opening to avoid condensation
  • →Handle in accordance with institutional biosafety and chemical handling guidelines
