Melanotan 1 vs Melanotan 2: The Key Differences Explained

A plain-English guide to how these two research peptides differ in shape, targets, handling, and lab use — written for Canadian researchers.

The difference between Melanotan 1 vs Melanotan 2 comes down to one thing: Melanotan 1 is a long straight chain that hits one target. Melanotan 2 is a short ring that hits four. Everything else follows from that.

The Short Answer

Neither one is "better." They are different tools for different jobs.

Picture a piece of string and a piece of string tied into a tight loop. The long flexible string can lay across a wide surface and grip it in many places. The small stiff loop cannot do that, but it slots neatly into lots of different openings.

Melanotan 1 is the string. Melanotan 2 is the ring. If switching on extra targets would ruin your result, use Melanotan 1. If comparing several targets is the point, use Melanotan 2.

People often describe these two as versions of the same thing. They are not. They come from the same starting point and share two of the same changes, but they are built in completely different shapes — and that changes what each one binds to, how it behaves in a test tube, how easily it dissolves, and which lab questions it can honestly answer.

The rest of this guide explains all of it in plain terms, with no chemistry background needed.

First, What Is a Peptide?

A peptide is a short chain of amino acids. Amino acids are the small parts that living things use to build proteins. If a protein is a long sentence, a peptide is a short phrase.

Because peptides are small and simple, scientists use them to study how the body sends and receives signals. They are studied in labs, in test tubes and cell samples, not given to people. If you are brand new to this, our guide on what research peptides are is a good place to start.

Both Melanotan 1 and Melanotan 2 are peptides. Both are sold in Canada for lab research only.

Both Come From the Same Natural Signal

The body makes its own signal peptide called alpha-MSH. The letters stand for alpha-melanocyte-stimulating hormone, but the name does not matter much. What matters is what it does: it tells certain cells to switch on.

Alpha-MSH is 13 building blocks long. Scientists wanted to study it, but it has two big problems.

  • It breaks down fast. In a lab sample, alpha-MSH falls apart in a few minutes. That is far too quick for most tests.
  • One of its parts rusts. The building block at position 4 is called methionine, and it reacts with oxygen. So a bottle of alpha-MSH slowly turns into a mix of good peptide and damaged peptide.

Both Melanotan compounds were made to fix these two problems. And here is the part most articles skip: they fix them the exact same way.

Fix 1: Swap the part that rusts

Scientists replaced the methionine at position 4 with a similar part called norleucine. Norleucine is almost the same size and shape, but it has no sulfur in it. Sulfur is the part that reacts with oxygen. So the rusting problem goes away, and the peptide still fits its target.

Fix 2: Flip one part backwards

Amino acids can exist in two mirror-image forms, like a left hand and a right hand. Living things almost always use the "left" form. So enzymes that chop up peptides are only trained to recognize left-handed parts.

Scientists flipped the phenylalanine at position 7 into its "right-handed" version, written as D-Phe. Two good things happened. The chopping enzymes no longer recognized that spot, so the peptide lasted much longer. And the flip locked a key section into the exact bent shape the target likes, which made it bind far more strongly.

Why This Matters

These two swaps were first described by Sawyer and his team in 1980. Their paper called the result a peptide with "ultralong biological activity." Later work found it binds around 1,000 times more strongly than the natural signal it came from. That is not a small tweak. That is the whole reason these compounds exist.

What Melanotan 1 Actually Is

Here is the simple version: Melanotan 1 is alpha-MSH with those two parts swapped, and nothing else changed.

It is still 13 building blocks long. It is still a straight chain. It still looks almost exactly like the natural signal. It just does not fall apart and does not rust.

Melanotan 1 also has an official medical name, afamelanotide. You may see it written as MT-1. All three names mean the same molecule.

Our Melanotan 1 research overview goes into the two substitutions, the receptor profile and the published record in more depth.

Because it kept the full natural length, Melanotan 1 is the closest stand-in scientists have for the real thing. When a study needs a signal that behaves like the body's own version but survives long enough to measure, this is usually the pick.

What Melanotan 2 Actually Is

Melanotan 2 came about ten years later, from a team at the University of Arizona. They took a completely different approach.

Instead of leaving the chain long, they did two things.

First, they cut it down. Earlier research had figured out that only the middle section, roughly parts 4 through 10, carries the actual message. Everything else was extra. So they threw the extra away. Thirteen building blocks became seven.

Second, they closed it into a ring. They built a chemical bridge connecting one end of the short chain to the other. This turns a floppy string into a stiff loop. Scientists call this bridge a lactam, and this ring shape is what makes Melanotan 2 what it is.

The Melanotan 2 research overview covers the cyclisation and its receptor profile in detail.

Back to the string and the ring

This is where the comparison from the top of the page comes from. Melanotan 1 kept the full chain, so it stayed a long flexible string. Melanotan 2 was cut short and closed into a stiff loop.

A long flexible string can lay across a big surface and grip it in many places at once. A small stiff ring cannot do that, but it slots neatly into lots of different small openings. That one difference explains almost everything else about how these two behave.

The Five Targets, and Why Focus Matters

Cells have parts on their surface called receptors. A receptor works like a lock. A peptide works like a key. When the right key goes into the right lock, the cell does something.

There are five locks in this family. Scientists call them MC1R through MC5R. They sit in different places and do different jobs.

ReceptorWhere it mostly sitsMelanotan 1Melanotan 2
MC1RPigment cells and some immune cellsStrongStrong
MC2RAdrenal glandVery weakVery weak
MC3RBrain and nervous systemWeakStrong
MC4RBrain and nervous systemWeakStrong
MC5RGlands and other tissueWeakStrong

Look at the last two columns and the difference jumps out. Melanotan 1 turns on one lock strongly and barely touches the rest. Melanotan 2 turns on four of them.

Now, why does that matter in a lab?

Say a researcher wants to know what happens when MC1R switches on. If they use Melanotan 1, and something changes, that change almost certainly came from MC1R. Clean answer.

If they use Melanotan 2 instead, they just switched on four locks at once. Something changed, but which lock caused it? They cannot say. The result is muddy.

The Simple Rule

If turning on extra targets would ruin your answer, use Melanotan 1.

If you are trying to compare several targets at once, use Melanotan 2.

Using Melanotan 2 for an MC1R-only question is the most common mistake in this area, and it makes the data hard to defend.

Melanotan 2 and PT-141 Are Nearly the Same Molecule

This trips up a lot of people, so it is worth clearing up.

PT-141, also called bremelanotide, is not a separate discovery. It is Melanotan 2 with one small chemical group swapped at the tail end. That is the entire difference.

You can see it in the numbers. Melanotan 2 weighs about 1,024. PT-141 weighs about 1,025. They are one unit apart.

So the family tree looks like this. Natural alpha-MSH sits at the top. Melanotan 1 is the branch that kept the full chain. Melanotan 2 is the branch that got cut short and looped. PT-141 sits right next to Melanotan 2 on that second branch.

Melanotan 1 is not closely related to either of them beyond that shared starting point. See the PT-141 overview for that branch of the family.

There is a fourth relative worth knowing. KPV is the final three residues of alpha-MSH — Lys-Pro-Val — studied on its own, separate from the melanotropic activity the full sequence carries. Where Melanotan 1 keeps the whole molecule and Melanotan 2 keeps the middle, KPV keeps only the tail. Our KPV overview covers it.

Storage and Handling: What Actually Changes

Both compounds arrive as a freeze-dried powder. Freeze-dried, sometimes called lyophilized, just means all the water was removed. A dry powder lasts far longer than a peptide sitting in liquid.

Most of the storage rules are the same for both. A few things differ.

PointMelanotan 1Melanotan 2
Dissolving in waterEasier. The long chain has no tight greasy centre.Harder. The ring is more compact and water-resistant.
Rusting riskRemoved by the norleucine swapRemoved by the same swap
Light damageYes — both contain tryptophan, which reacts to lightSame
Extra thing to check on the COANothing special. No ring, no sulfur bridges. The ring must be intact. A broken ring behaves differently.
Metal in buffersCan bind zinc and copper — avoid metal buffersSame

General rules that apply to both: keep the sealed powder cold and dry, keep it out of light, and once it is mixed with liquid, split it into small portions instead of freezing and thawing the same vial over and over. Every freeze-thaw cycle damages a little more of it.

Our guide to peptide stability covers temperatures, degradation routes and timelines in detail, and why some peptides reconstitute faster than others explains the solubility difference described above.

How to Check You Are Getting the Real Thing

Here is a problem with both of these compounds. They look identical in the vial. A white powder is a white powder. You cannot tell a 99% pure batch from a 70% pure batch by looking at it, and you cannot tell Melanotan 1 from Melanotan 2 either.

The only way to know is paperwork. Specifically, a Certificate of Analysis, usually shortened to COA.

A COA is a lab report for that exact batch. Here is what to look at.

  • Purity. Should be 99% or higher. If no number is given, that is a bad sign.
  • Test method. Look for HPLC and Mass Spectrometry. HPLC measures how much of the sample is the real compound. Mass Spectrometry confirms it is actually the compound they claim, by weighing the molecule. You want both.
  • Molecular weight. This is a quick sanity check you can do yourself. Melanotan 1 should come in around 1,647. Melanotan 2 should come in around 1,024. If a COA labelled Melanotan 1 shows a weight near 1,024, something is wrong.
  • Who tested it. An outside lab, not the seller's own lab. A company grading its own homework is not proof of anything.
  • Batch number. The COA should match the exact batch you received, not be a generic file reused for every order.

For Melanotan 2 there is one extra check. Because the molecule is a closed ring, and rings can break open during manufacturing, a good COA confirms the ring is intact. Ring-opened material is a different compound that behaves differently.

You can view NexPept's third-party COAs on our peptide COAs page.

Buying Melanotan 1 or Melanotan 2 in Canada

Both are sold in Canada as research compounds for laboratory use only. Neither is approved for use in people or animals. Our guide on peptide rules in Canada covers how this works in detail.

When comparing Canadian suppliers, the same things matter for these two as for any research peptide.

  • Outside lab testing with a COA you can actually see before you buy
  • Batch tracking, so the vial in your hand matches a specific report
  • Clear research-use-only labels on the product, with no dosing or human-use claims anywhere
  • Domestic shipping, which avoids customs delays and long trips at uncontrolled temperatures
  • Real support, meaning someone who can answer a storage or handling question properly

Both are listed in our catalogue: Melanotan-1 10mg and Melanotan-2 10mg, each batch-tested with its certificate published before you order.

Shipping matters more than people expect with peptides. A package sitting in customs for two weeks is two weeks of temperature swings. Our page on how we ship peptides in Canada explains our process.

Common Mistakes Researchers Make

Treating them as interchangeable

They are not. Swapping one for the other changes which receptors get switched on. If a protocol says Melanotan 1, using Melanotan 2 instead does not just change the strength of the result. It changes what the result means.

Assuming Melanotan 2 is "stronger"

Broader is not the same as stronger. At MC1R, both are powerful. Melanotan 2 simply hits more targets. Calling it stronger is like calling a wide paintbrush stronger than a fine one.

Skipping the COA

Two white powders. No way to tell them apart by eye. The paperwork is the only thing standing between a clean experiment and weeks of confusing data.

Repeated freeze-thaw

Mixing a whole vial, freezing it, thawing the whole thing for every use, and refreezing it will slowly wreck the sample. Splitting it into small single-use portions the first time is a five-minute job that protects the whole batch.

Ignoring light

Both contain tryptophan, which is damaged by light. A clear vial on a lit bench is losing quality while you work. Amber vials or foil solve it.

Melanotan 1 vs Melanotan 2: The Short Version

  • Both come from the same natural signal, alpha-MSH, and share the same two fixes
  • Melanotan 1 is a long straight chain of 13 parts; Melanotan 2 is a short ring of 7
  • Melanotan 1 focuses on one receptor, MC1R; Melanotan 2 hits four
  • Neither is better — it depends on whether you need a precise tool or a broad one
  • PT-141 is nearly identical to Melanotan 2, not to Melanotan 1
  • Melanotan 1 dissolves more easily; Melanotan 2 needs its ring checked on the COA
  • Both are sold in Canada for laboratory research only

Common Questions About Melanotan 1 vs Melanotan 2

What is the main difference between Melanotan 1 and Melanotan 2?

Shape and focus. Melanotan 1 is a long straight chain of 13 building blocks that mainly switches on one receptor, MC1R. Melanotan 2 is a short ring of 7 building blocks that switches on four receptors. They are different molecules, not two strengths of one.

Is Melanotan 2 stronger than Melanotan 1?

Not in the way people usually mean. At MC1R, both bind powerfully. Melanotan 2 is broader, not stronger. It reaches more receptor types, which is useful in some studies and a problem in others.

Why does Melanotan 1 have a second name, afamelanotide?

Afamelanotide is the official international name for that molecule. Melanotan 1 and MT-1 are the informal names used in research and by suppliers. All refer to CAS 75921-69-6. Having an official name does not mean research-grade material is the same as any approved medicine — research material is made and tested to laboratory standards, not pharmaceutical ones.

What is the difference between Melanotan 2 and PT-141?

Very little. PT-141, also called bremelanotide, is Melanotan 2 with one small chemical group changed at the end of the molecule. Their weights differ by about one unit. They are close relatives. Melanotan 1 is not.

Which one is used more in published studies?

It depends on the field. Melanotan 1 appears more in pigment cell and light-protection research, where a single clean target matters. Melanotan 2 appears more in receptor-mapping and brain pathway work, where reaching several targets is the goal. Both show up often in structure studies, because comparing a string to a ring is a useful lesson.

Do they need different storage?

Mostly no. Keep the sealed powder cold, dry and dark. Once mixed with liquid, split into small portions and avoid repeated freezing and thawing. The main difference is on paper rather than in the freezer: Melanotan 2 has a ring that should be confirmed intact on the Certificate of Analysis.

Why does Melanotan 2 dissolve less easily?

Its ring shape packs the water-repelling parts of the molecule into a tight centre. Melanotan 1, being a long open chain, spreads those parts out, so water gets to it more easily.

Can I tell them apart by looking at the vial?

No. Both are white or off-white freeze-dried powder and look identical. The only reliable check is a batch-specific Certificate of Analysis showing purity, test method, and molecular weight. Melanotan 1 lands near 1,647 and Melanotan 2 near 1,024, so the weight alone will tell you which one you have.

What does "lyophilized" mean?

It means freeze-dried. The water was frozen and then removed, leaving a dry powder. Peptides last much longer dry than they do sitting in liquid, which is why they are shipped and stored this way.

Are Melanotan 1 and Melanotan 2 legal in Canada?

Both are supplied in Canada for laboratory research use only, with research-use-only labelling. Neither is approved for human or veterinary use. Our Canadian peptide regulations guide covers the details.

What purity should I look for?

99% or higher, confirmed by HPLC and Mass Spectrometry, tested by an independent lab rather than the seller. The report should be tied to the specific batch you receive, not a generic file.

Final Thoughts

The easiest way to hold onto the difference is the string and the ring.

Melanotan 1 is the string. Long, flexible, close to what the body already makes, and focused on one target. It is the tool you reach for when you need a clean answer about one thing.

Melanotan 2 is the ring. Short, stiff, compact, and able to fit into several different targets. It is the tool you reach for when you want to see across the whole family at once.

Both are useful. Neither replaces the other. And with either one, the batch paperwork is what turns a white powder into a reliable experiment.
