For laboratory research use only. Not for human or veterinary use.

Melanotan-1 is a 13-residue synthetic analogue of alpha-melanocyte-stimulating hormone, distinguished from the natural hormone by exactly two amino acid substitutions. Those two changes are the entire story of the compound: they make it far more stable and far more selective than the peptide it was derived from.

What is Melanotan-1?

Melanotan-1 — known in pharmaceutical contexts as afamelanotide, and in the literature by the designation [Nle4, D-Phe7]-α-MSH or NDP-MSH — is a synthetic peptide built on the backbone of alpha-melanocyte-stimulating hormone. It was synthesised at the University of Arizona in the 1980s and later licensed to an Australian company for clinical development.

Natural α-MSH is a 13-residue hormone that signals through the melanocortin receptor family. It has an obvious practical problem as a research tool: it degrades quickly, and it binds several melanocortin receptor subtypes rather than one. Melanotan-1 was designed to address both.

The two substitutions, and why they matter

This is the part worth understanding properly, because it explains both why the compound exists and how to handle it.

Norleucine at position 4

Natural α-MSH carries methionine at position 4. Methionine contains sulfur, and that sulfur is readily oxidised — it is consistently among the most chemically fragile residues in any peptide sequence. Oxidation changes the molecule's mass and can change its behaviour, which makes a methionine-containing peptide awkward to store and to work with reproducibly.

Norleucine is a near-perfect structural stand-in: the same chain length and shape, with the sulfur atom replaced by a carbon. The molecule keeps its geometry and loses its most vulnerable chemical liability. This is a recurring strategy in peptide design, and it is why Melanotan-1 does not carry the same oxidation caution that sequences like Semax or MOTS-c do.

D-phenylalanine at position 7

The second substitution swaps the natural L-form of phenylalanine for its mirror image, the D-form. Almost all amino acids in naturally occurring proteins are L-form, and the enzymes that break peptides down are built to recognise that geometry. A D-residue is, to those enzymes, the wrong shape — so it resists cleavage.

The change also locks the local backbone into a conformation that happens to improve receptor binding. Published sources describe Melanotan-1 as binding the MC1 receptor for considerably longer than natural α-MSH does. One substitution therefore buys both durability and affinity, which is unusual; most stability modifications cost some potency.

Why this is worth reading closely: a great deal of peptide design consists of exactly these two moves — replace an oxidation-prone residue with a stable isostere, and introduce a D-amino acid or an unnatural residue where proteases cut. Melanotan-1 is an unusually clean textbook case because the parent hormone is well characterised and only two positions changed, so what each substitution contributes can be attributed rather than guessed at.

Receptor selectivity: the key difference from Melanotan-2

The melanocortin receptor family has five known subtypes. MC1R is associated with pigmentation signalling; the others are distributed differently and are associated with different systems. Melanotan-1 binds predominantly to MC1R.

Melanotan-2 is a different molecule — shorter, cyclic, and considerably less selective, binding several melanocortin subtypes. For experimental purposes that difference is the whole point: a selective ligand lets you attribute an observed effect to one receptor, while a non-selective one does not. Researchers often use both precisely to exploit that contrast.

We cover the comparison in detail on a dedicated page: Melanotan-1 vs Melanotan-2, which sets the two sequences, receptor profiles and research applications side by side. If you are deciding which of the two suits a given study design, start there rather than here.

A structural note: the KPV connection

Look at the last three residues of the Melanotan-1 sequence: Lys-Pro-Val. Those same three residues close out natural α-MSH, and on their own they are the tripeptide known as KPV — α-MSH(11–13).

This is not a coincidence, and it is a useful thing to know when reading across this literature. KPV is studied for signalling behaviour attributed to that short C-terminal fragment alone, separate from the melanotropic activity the full sequence carries. Melanotan-1 and KPV therefore sit at opposite ends of the same investigative question: what does the whole α-MSH molecule do, and how much of it is actually required?

PT-141 belongs to the same family from a third direction — it is a metabolite of Melanotan-2 rather than of α-MSH directly, with its own receptor profile.

Research contexts

Published work involving Melanotan-1 centres on melanocortin receptor pharmacology, where its selectivity makes it a standard reference ligand for MC1R, and on pigmentation signalling, since MC1R is the subtype most closely associated with melanin pathways in model systems.

Beyond that, it appears regularly in structure–activity work, where its well-defined relationship to the parent hormone makes it a useful comparator, and in peptide stability studies, where NDP-MSH is frequently used as an example of successful stabilisation. In our catalogue it sits under neuro research, alongside the other melanocortin compounds.

Regulatory status

Afamelanotide — the pharmaceutical form of this peptide — is an approved prescription medicine in some jurisdictions. It received European approval in January 2015 and United States FDA approval in October 2019, under the brand name Scenesse, for a rare inherited photosensitivity disorder called erythropoietic protoporphyria. It was designated a first-in-class medication.

Important regulatory context Those approvals are for a specific pharmaceutical product, for one rare disease, manufactured under pharmaceutical controls and administered under medical supervision. They confer nothing on research-grade material and they are not Health Canada authorisations. Melanotan-1 supplied by NexPept is not a drug, not a supplement, not a cosmetic and not a treatment for any condition. It is intended for laboratory research use only and is not for human or veterinary use.

Handling and storage in the laboratory

Melanotan-1 is supplied as a lyophilised powder, stable frozen and markedly less so once reconstituted. Standard practice is to keep sealed vials frozen and protected from light and moisture, and to bring a vial to room temperature before opening so that atmospheric moisture does not condense inside it.

The sequence has a useful property here. Because position 4 carries norleucine rather than methionine, this peptide is not subject to the methionine-oxidation route that complicates many others, so the usual warning about minimising air exposure applies with less force. It does contain tryptophan at position 9, which is photosensitive, so light protection remains genuinely important rather than boilerplate.

Reconstitution solvent, working concentration and aliquoting strategy follow the protocol of the study being run. Avoid repeated freeze-thaw cycling, and record lot numbers against experiments so a batch change midway through a series does not become an undiagnosable confound later.

Verification and certificates of analysis

Every Melanotan-1 lot NexPept lists is tested by an independent third-party laboratory using high-performance liquid chromatography and mass spectrometry, and the report is published openly rather than offered on request after purchase. Certificates are in the NexPept Certificate of Analysis library, with the batch number on each vial matching a published report. From late September 2026 we have also been adding heavy-metal and endotoxin testing to those reports.

When reading any certificate, confirm the batch number matches your vial, that both an identity method and a purity method are reported, that the test date is recent, and that the issuing laboratory is named. Our quality standards page covers how batches are tested and documented, what research peptides are explains how research-grade material differs from a drug or supplement, and buying peptides in Canada walks through what to check before ordering. The broader guide to research peptides in Canada covers sourcing and regulation.

Frequently asked questions

What is the difference between Melanotan-1 and Melanotan-2?

Different molecules, not two versions of one. Melanotan-1 is a 13-residue linear analogue of α-MSH that binds predominantly to MC1R. Melanotan-2 is shorter, cyclic, and binds several melanocortin receptor subtypes. The selectivity difference is the main experimental distinction — our Melanotan-1 vs Melanotan-2 comparison sets them out side by side.

Why is it also called afamelanotide and NDP-MSH?

Three names for one compound. Melanotan-1 is the common research name; afamelanotide is the international nonproprietary name used in pharmaceutical contexts; NDP-MSH, or [Nle4, D-Phe7]-α-MSH, is the structural designation describing the two substitutions. Searching the literature is more productive if you try all three.

What do the two amino acid substitutions actually do?

Norleucine at position 4 replaces methionine, removing the sulfur atom that makes methionine prone to oxidation, without changing the residue's shape. D-phenylalanine at position 7 replaces the natural L-form; proteases are built to recognise L-amino acids, so the mirror-image residue resists cleavage, and it also improves receptor binding. Together they make the molecule more stable and longer-binding than the hormone it derives from.

How does Melanotan-1 relate to KPV?

KPV is the final three residues of this sequence — Lys-Pro-Val, α-MSH(11–13). It is studied for behaviour attributed to that fragment alone, separate from the melanotropic activity the full sequence carries. See the KPV overview for that side of the literature.

Is Melanotan-1 approved in Canada?

No. Afamelanotide is approved in the EU and United States under the brand name Scenesse for a rare photosensitivity disorder, but there is no Health Canada authorisation, and an approval elsewhere confers nothing here. Research-grade material is not a drug, supplement or cosmetic, and is supplied for laboratory research use only.

Does this peptide need the same oxidation precautions as others?

Less so, and for a specific reason: the methionine that would normally be the oxidation-prone residue has been replaced by norleucine. Light protection still matters because of the tryptophan at position 9. Otherwise the usual practice applies — frozen, sealed, brought to room temperature before opening, aliquoted to avoid freeze-thaw cycling.

What purity should I expect, and how is it verified?

NexPept lists Melanotan-1 at ≥99% purity, confirmed by HPLC and mass spectrometry before a lot is listed. Reports for every batch we have shipped are published in our COA library, so the certificate is available before you order rather than after.
